I would go tuner > eq (although this could be useful anywhere) > overdrive > wah > delay > noise suppressor > amp. You might find that you can get good results with some of the stuff in the effects loop, but personally I probably wouldn't stray far from that order.

The other guitarist in my band has a mesa nomad, a 50watt 1 x 12. He has been having reliability problems with it but it's old, and the high gain sound is great.

I don't know much about interfaces, but I have a focusrite saffire 10 i/o, they also do a smaller interface with 4 xlr inputs which would be perfect for solo vocal and guitar.
